Abstract : Background: Liraglutide is a Glucagon-like-peptide analogue that is used for the management of high blood glucose levels in type 2 diabetes mellitus and for weight reduction in obesity. In addition to its anti-inflammatory qualities, intriguing emerging research demonstrated that liraglutide can overcome brain metabolism changes associated with aging and Alzheimer disease, such as neuronal damage. Emerging research recommend that liraglutide exerts its effects through inhibiting the production of amyloid beta, boosting synaptic plasticity, and increasing brain tissue glucose uptake. liraglutide was combined with memantine in this study to assess its efficacy in managing experimental induced AD in the animal model and slowing its progression. Methodology: The study comprised 50 male Wistar rats, which were divided into five groups of ten animals each: control positive, control negative, memantine treated group, liraglutide treated group, and memantine + liraglutide treated group. The disease-inducing agents were aluminum chloride + d-galactose, and the study lasted ten weeks. Results: The combination regimen experimental results showed remarkable reduction in beta amyloid protein level in the brain tissue. In addition, glucose level was well controlled when combination treatment was used, and glycemic management was improved. As a result of reduced amyloid beta protein level neuronal inflammation and oxidative stress was reduced showing improved levels of antioxidant parameters. Moreover, the Morris Water Maze test (MWM) indicated enhanced outcomes in combination treated group by showing the shortest latency among all treated groups. The combined effects of liraglutide + memantine most likely showed promising therapy in controlling Alzheimer disease symptoms. Conclusion: The combination of liraglutide + memantine showed neuroprotective effects and enhanced memory and cognitive functions in this experimental animal model of Alzheimer disease. key words: Alzheimer disease, cognitive function, liraglutide, memantine, glucagon-like peptide-1
