Abstract : The immune system has the ability to protect the body from attacking pathogens by removing these pathogens. Natural killer cells are among the most influential components of the immune system. The main important function of NK cells is the ability to recognize and remove cells that do not have such as in the case of viral infection or cancer cells. Typically MHC class I molecules, people use natural products when they get infections in an effort to avoid the side effects of medications. The black seed is one of the most important natural products that people have used since ancient times in treatment. It has an important role as an anti-TQ and one of the most important components of the black seed. TQ has been shown to be popular, as it is an anti-inflammatory and antitumor TQ on cells. We are interested in this study to look at the effect on the cytotoxic capacity of natural killer TQ. This will be done by analyzing the effect of NKG2D and NKp by studying the expression of 30 natural killer cell activation molecules. Mononuclear blood cells will be isolated and then normal cells will be isolated. B cytokines and lethal granzyme secretion. The study will be conducted on five different cultures of natural killer cells (untreated), killer cells treated with thymoquinone (treated), cancer cells alone, untreated killer cells cultured with tumor cells and killer cells treated with thymoquinone with cancer cells. After the supernatant is collected, cytokines will be detected using ELISA. Cells are detected by a flow cytometer. Finally, surface NKP30,NKG2D secretion of granzyme B by NK cells will be detected in the supernatant using ELISA. No detectable changes in the effect of thymoquinone on natural killer cells were recorded for all data. In conclusion, thymoquinone did not show any effect on NK cells or their cytotoxic capabilities under the conditions of the experiment that was performed and more detailed studies with more conditions proposed can be conducted to support this study.
