Abstract : Aim: Advanced glycation end products (AGEs) play a major role in the development of several vascular complications that are mediated by endothelial dysfunction. The present work aimed to investigate the mechanism by which AGEs impair vascular relaxation. Methods: The effect of AGEs on vasodilation induced by acetylcholine (ACh) or D NONOate was examined by incubating rat aorta with different concentrations of methyle glyoxal (MG) an AGEs producer compound. ACh-induced nitric oxide(NO)generation, a potent vasodilator was assessed using the fluorescent probe diaminofluorecein (DAF-FM). In addition, The effect of AGEs on mRNA expression of arginase 2 (Arg2), nicotinamide adenine dinucleotide phosphate (NADPH) oxidase (Nox4) and endothelial nitric oxide synthase (Nos3) was determined by quantitative real-time polymerase chain reaction(qRT- PCR). Results: One-hour in vitro incubation of rat aorta with AGEs impaired endothelial-dependent vasodilation produced by ACh, while D NONOate-induced vasodilation has increased. Pre-incubation of aorta with ornithine, an arginase2 inhibitor prevented the vasodilation impairment effect induced by AGEs. In addition, tempol, a superoxide scavenging or apocynin, a NADPH oxidase inhibitor blocked the effect of AGEs. ACh-induced NO production was decreased by AGEs and this was inhibited by both ornithine and apocynin. Furthermore, AGEs exposure increased Arg2 mRNA expression but decreased Nos3 mRNA expression in isolated rat aorta. Conclusion: The present study indicates that AGEs impaired endothelial-dependent vasodilation,and this effect is mediated by arginase overexpression and NADPH oxidase stimulation.
