EFFECT OF MELATONIN ON ISCHEMIC-REPERFUSED RAT HEARTS

Abstract : Aim: This work was planned to investigate the probable protection by melatonin supplementation on cardiac vulnerability to ischemia and reperfusion Subjects and Methods: Rats were allocated into three groups, I- Melatonin-treated group in a small dose (n= 10), melatonin was given in a dose of 5mg/kg/day for 14 consecutive days, orally. II- Melatonin-treated group in a high dose (n= 10), melatonin was given in a dose of 10 mg/kg/day for 14 consecutive days, orally. III¬Control group (n= 10) was treated with distilled water (solvent of melatonin) orally. Hearts of rats in all groups were subjected to 30 min. of ischemia followed by 45 min. of reperfusion in vitro. Results: The present study demonstrated that administration of melatonin in small and high dose in normal rats produced significant increase in the basal peak developed tension (PT) associated with significant shortening of time to peak tension (FPT) and half relaxation time (112 RT). In addition, there was a significant increase in the basal heart rate (HR) and coronary flow rate (CFR ) particularly in high dose treated group. Also, melatonin administration showed an improvement in the cardiac performance during post-ischemia reperfusion period, manifested as, significant increase in the percentage of baseline values in PT, TPT, and 112 RT, associated with significant increase in the percentage of HR and CFR. Myocardial cellular integrity, estimated by plasma level of creatine kinase(CK) and lactate dehydrogenase (LDH) was improved by melatonin administration, revealed evidently by the significant decrease in CK and LDH release in the coronary effluent of rats treated with melatonin. Conclusion: Melatonin has a beneficial cardio-protective effects and prevents the decrimental effects of postischemia reperfusion on the myocardial contractility.
